Stage
1
Insertng Microorganisms
Syringe your bacterial solution (1-1.5ml) into the BioCapsule.
To do so, pierce the BioCapsule at it’s top. Handle this stage carefully to avoid needle injury.
Please avoid suspension spillage on capsule surface.
Stage
2
Sealing
Open the vial included in the kit and draw 5 μƖ of the sealing polymer using a pipette. Carefully release the sealing polymer onto the needle hole.
Stage
3
Drying
Dry the sealing polymer for 5 minutes,
preferably within a biological hood.
Before the sealing procedure
After the sealing procedure
Stage
4
Repeat Sealing
Repeat stages 2 and 3 another two times.
Note that the sealing polymer should dry and change its color to white.
Sterilize the capsule surface with a 70% ethanol solution for 3 seconds.
Stage
5
Capsule Activation
Incubate the filled BioCapsules within saline and nutrients (2%) or other suitable medium for at least 72 hours.
Best incubated in an orbital shaker.
Once the Capsules are activated they will sink to the bottom of the flask or float veridically.
Special notes:
- The capsules are sterile as long as the capsule bag was not opened. The polymer is sterile whilst the vial is sealed. To keep sterile after opening, handle in a biological sterile area such as a biological hood.
- Activated capsules should not be extracted from the medium for more than 10 seconds.
- The BioCapsules are acitve when their position within the incubated medium is vertical
or if they sink to the bottom of the incubation vessel.
